Course Description:

Lissencephaly is a rare brain malformation associated with mental retardation and epilepsy that can occur as an isolated abnormality or in association with various syndromes. The condition is characterized by absent (agyria) or decreased (pachygyria) convolutions of the cerebral cortex, causing the brain's surface to appear smooth. Lissencephaly is generally divided into two categories: classic lissencephaly (also known as type 1) and cobblestone lissencephaly (also known as type 2). Children with classic lissencephaly often appear normal as newborns but may have hypotonia, poor feeding, and apnea. They tend to exhibit signs of failure to thrive early and develop spasticity and seizures within the first year. Cobblestone lissencephaly is often associated with features that are not typically seen in classic lissencephaly, including hydrocephalus, eye abnormalities, muscular dystrophy, and cerebellar hypoplasia. Other lissencephaly syndromes, some that include multiple congenital anomalies, have also been described. Lissencephaly manifestations are variable in nature, requiring development of an individualized treatment plan that takes each patient’s unique circumstances into account. Therapy focuses on maximizing potential, preventing secondary complications, providing supportive care for comfort, and educating caregivers.
